The CD40/NFκB Reporter Lentivirus is designed to study the signaling and activation of CD40 in antigen-presenting cells (APCs). The construct constitutively expresses the CD40 receptor, which activates APCs such as B cells, macrophages, and dendritic cells upon engagement. This interaction triggers NFκB-dependent transcription and the expression of dual reporters: secreted Gaussia luciferase (GLuc) and GFP. These reporters allow researchers to monitor CD40-mediated signaling and immune activation in real time. Example applications include assessing APC functionality, studying immune regulation, and testing CD40-targeting therapeutics. To use this system, transduce your desired cells with the all-in-one high-titer purified lentivirus to establish a stable, pathway-specific cell line for immunological studies. This product is supplied as high-titer lentiviral particles (third generation), purified by PEG precipitation and sucrose gradient centrifugation. The lentiviral particles are VSV-G pseudotyped and are ideal for difficult-to-transfect cells, including primary or thawed cells.
