The CTLA4/NFAT Immunotherapy Reporter Lentivirus is designed to study the inhibitory effects of CTLA4-mediated immune checkpoint signaling in T cells. The construct constitutively expresses the CTLA4 receptor, which binds to B7 ligands on antigen-presenting cells, leading to suppression of T cell activity. This engagement decreases cytosolic Ca2+ levels, reducing NFAT activation and driving a corresponding decrease in reporter output: secreted Gaussia luciferase (GLuc) and GFP. Researchers can also use this system to study receptor engagement inhibition, such as with anti-CTLA4 antibodies, which restore reporter activity by relieving CTLA4-mediated suppression. These reporters enable real-time monitoring of CTLA4 signaling and therapeutic interventions that modulate immune checkpoint activity. To use this system, transduce your desired cells with the all-in-one high-titer purified lentivirus to establish a stable, pathway-specific cell line for immunotherapy and immune checkpoint studies. This product is supplied as high-titer lentiviral particles (third generation), which have been purified by PEG precipitation and sucrose gradient centrifugation. The lentiviral particles are VSV-G pseudotyped and are ideal for difficult-to-transfect cells, including primary or thawed cells.
