The GITR/NF-κB Immunotherapy Reporter Lentivirus is designed to measure the co-stimulation effect of GITR on T-cell activation. Upon binding by an agonist antibody or GITR ligand expressed on target cells, GITR on effector cells activates the NF-κB signaling pathway, enhancing T cell activation and immunity against target cells, such as tumor cells. This reporter lentivirus is an efficient tool for generating stable, sensitive T-lymphocyte lines to screen for GITR agonists.
The construct features constitutive expression of GITR and a drug selection marker (BSD), driven by the EF1a promoter, along with a dual reporter system comprising secreted Gaussia luciferase (GLuc) and GFP, driven by NF-κB response elements. This all-in-one high-titer purified lentivirus simplifies the process of establishing stable cell lines for functional assays.
This product is supplied as high-titer lentiviral particles (third generation), purified by PEG precipitation and sucrose gradient centrifugation. The lentiviral particles are VSV-G pseudotyped, ideal for transducing difficult-to-transfect cells, including primary or thawed cells.

T cell Activation. The product can be used to generate stable cell lines that read out GITR signaling through downstream activity of NFκB, for example in stable T or NK cell lines. The fluorescent and luminescent dual reporter provides flexibility in the readout approach. GLuc is secreted, allowing the user to detect it in various biological media. Generally, the product is useful for analyzing T cell activity as a function of GITR stimulation/inhibition. |

Figure 1. GITR/NFκB Reporter Fluorescence Validation HEK293FT cells were transfected with GITR/NFκB Reporter construct (EF1α-GITR-P2A-GFP, NFκB-RFP-GLuc). Constitutively expressed GITR and GFP are shown in the left and middle panels respectively, while RFP fluorescence in the right panels shows activation of the NFκB reporter in response to stimulation with human TNFα. The arrangement of components in this unique reporter system allow the user to select transduced cells by FACS sorting of GFP+ cells, whereas other variations of the product utilize a drug selection marker instead. Besides RFP expression, NFκB pathway activation can be examined also by luminescence detection of secreted Gaussia luciferase, depending on the user’s application or available instrumentation. Can I purchase the plasmid for this product?
TF reporter products such as this one are not available in plasmid format. They are only available as high-titer lentiviral particles. Other gene products on our site, such as ORF cDNA and shRNA products are available in either format.
