ATF6 Reporter Cell Lines
Overview
Details
The ATF6 HEK293 Reporter Cell Lines collection is engineered to evaluate ATF6 signaling signaling activity with high precision. Produced by stable transduction with our lentiviral reporter systems, these HEK293 cells stably express a ATF6 responsive reporter construct, offered in a range of formats including fluorescent or luminescent reporters. Formats may vary by availability. Upon pathway activation, ATF6 regulates downstream transcriptional programs that are captured by the integrated reporter. These cell lines provide flexible, ready-to-use platforms for real-time analysis of pathway modulation, transcriptional activity, and small molecule screening.

Product Data:

Figure 1. ATF6-Dependent Activation in HEK293 mCherry Reporter Cells
HEK293 ATF6 mCherry Reporter Cells (Puro) were treated with Tunicamycin or Thapsigargin at the indicated concentrations for 36 h. Fluorescent imaging was performed to visualize ATF6-dependent reporter activation associated with ER stress.
Key Features:
- Reporter system:
- Fluorescent variants (e.g., eGFP, mCherry) enable real-time, live-cell monitoring of ATF6 signaling signaling, while bioluminescent variants (e.g., Firefly Luciferase) provide high-sensitivity, plate-based readouts compatible with traditional and high-throughput screening workflows.
- Stably integrated ATF6 responsive reporter construct driving reporter expression.
- Validated with to confirm pathway responsiveness.
- Ideal for live-cell imaging, flow cytometry, or luminescence-based assays depending on format.
Applications:
- Screening activators and inhibitors of the ATF6 signaling pathway.
- Studying transcriptional activity regulated by ATF6 signaling in mammalian systems.
- Evaluating ATF6-mediated transcriptional regulation.
- Fluorescence or luminescence-based readouts for high-throughput screening and mechanistic studies.
Quality Control: Each cell line is validated for reporter expression and pathway responsiveness using appropriate stimuli (e.g., ). Stable expression is confirmed for ≥20 passages. All cells are screened and certified mycoplasma-free.
Shipping and Storage: Shipped on dry ice (USA, Canada, Europe, Asia) Cells should be transferred to Liquid nitrogen vapor phase upon receipt.
How to Use: Plate cells and stimulate with the indicated pathway agonists or test compounds. Monitor reporter activity via fluorescence microscopy, flow cytometry, or luminescence detection depending on the construct.

Background:
ATF6 signaling signaling plays a central role in cellular homeostasis, development, and disease. Activation of this pathway leads to modulation of ATF6 activity and downstream transcriptional responses. The reporter system used in these cells leverages defined response elements and a minimal promoter to drive reporter expression, enabling sensitive and specific measurement of pathway activation. These HEK293 reporter lines offer robust tools for dissecting pathway-driven transcriptional responses and screening modulators in live cells.
