The TCF/LEF (Wnt/Beta-Catenin) HEK293 Reporter Cell Lines are optimized for sensitive, real-time monitoring of TCF/LEF-mediated Wnt/Beta-Catenin signaling signaling. Featuring a TCF/LEF responsive reporter construct driving fluorescent or luminescent reporter output, these cell lines enable direct analysis of TCF/LEF activity in live cells and are ideal for studying TCF/LEF-mediated Wnt/Beta-Catenin signaling pathway modulation and downstream transcriptional responses.

TCF/LEF (Wnt/Beta-Catenin) Reporter Cell Lines
Overview
Details
The TCF/LEF (Wnt/Beta-Catenin) HEK293 Reporter Cell Lines collection is engineered to evaluate TCF/LEF-mediated Wnt/Beta-Catenin signaling signaling activity with high precision. Produced by stable transduction with our lentiviral reporter systems, these HEK293 cells stably express a TCF/LEF responsive reporter construct reporter construct, offered in a range of formats including fluorescent or luminescent reporters. Formats may vary by availability. Upon pathway activation, TCF/LEF regulates downstream transcriptional programs that are captured by the integrated reporter. These cell lines provide flexible, ready-to-use platforms for real-time analysis of pathway modulation, transcriptional activity, and small molecule screening.

Product Data:
Figure 1. TOPFlash EGFP Reporter Activation by LiCl in Stable HEK293 Cells
TOPFlash EGFP reporter (RCL-0011-5S) stable HEK293 cells were generated using TCF/LEF-TAG-BSD lentiviral particles and selected with blasticidin. Cells were seeded at 5 × 104 cells per well in a 48-well plate and incubated for 8-10 hours. Cells were then treated with LiCl for 32 hours, trypsinized, and analyzed by flow cytometry to observe EGFP expression.
Figure 2. TOPFlash EGFP Reporter Activation by Human Wnt3a in Stable HEK293 Cells
TOPFlash EGFP reporter (RCL-0011-5S) stable HEK293 cells were seeded at 5 × 104 cells per well in a 48-well plate and incubated for 8-10 hours. Cells were treated with 2.5 mM LiCl for 16 hours before addition of human Wnt3a to a final concentration of 25 ng/ml, then incubated for an additional 16 hours. Cells were then trypsinized and analyzed by flow cytometry.

Background:
TCF/LEF-mediated Wnt/Beta-Catenin signaling signaling plays a central role in cellular homeostasis, development, and disease. Activation of this pathway leads to modulation of TCF/LEF activity and downstream transcriptional responses. The reporter system used in these cells leverages defined response elements and a minimal promoter to drive reporter expression, enabling sensitive and specific measurement of pathway activation. These HEK293 reporter lines offer robust tools for dissecting pathway-driven transcriptional responses and screening modulators in live cells.
