NeuroD1 Reporter Lentivirus: The NeuroD1 Reporter is a high quality lentiviral reporter system designed to measure the transcriptional activity of NeuroD1 in mammalian cells, using tandem repeats of NeuroD1 response elements within the enhancer of the secretin promoter. This setup allows for the detection of NeuroD1’s synergistic interaction with Sp1 and RREB1, proteins that are widely expressed and jointly enhance secretin gene transcription. NeuroD1 is a basic helix-loop-helix transcriptional factor, also known as BETA2, which is found in neurons, the anterior pituitary gland, pancreatic islets, and enteroendocrine cells, where it targets genes such as insulin, secretin, and pro-opiomelanocortin (POMC). Belonging to the neural lineage NeuroD family, which includes NeuroD1, NeuroD2, NeuroD4, and NeuroD6, NeuroD1 plays a pivotal role in cerebral cortex development and central nervous system regulation. It forms heterodimers with other transcription factors to control genes essential for these processes. Furthermore, NeuroD1 is crucial for β cell maturation and the expansion of pancreatic islet cell mass, underscoring its significance in both neural and endocrine system functions. The reporter lentivirus is purified by PEG precipitation and sucrose gradient centrifugation and is ideal for difficult-to-transfect cells including primary and/or thawed cells.

Product Data

Figure 1. GFP Reporter activation of NeuroD1-TAG-Puro by NeuroD1 HEK293FT cells were co-transfected with plasmids for NeuroD1-TAG-Puro along with empty vector (pcDNA) or cDNA encoding human NeuroD1 for 32 h before acquiring fluorescent microscopy images. NeuroD1 expression vector contains RFP downstream of NeuroD1, separated by P2A. This product is supplied as pre-packaged lentiviral particles with your choice of reporter and selection marker. Featured cDNA expression vectors are also available upon request. For more information, please email info@lipexogen.com.

Recommended Controls
Reporter Negative Control Lentivirus – Ready-to-transduce lentiviral particles expressing minimal TATA box-driven reporter. The construct is the same as the TF Reporters except that it lacks the transcriptional response elements which drive signal pathway/TF-specific reporter expression. The reporter negative control lentiviral particles allow to establish a baseline for background reporter activity and determine specificity of any treatments to activate the reporter.
Reporter Positive Control Lentivirus – Ready-to-transduce lentiviral particles with constitutive expression of the reporter. The reporter positive control lentivirus is useful for transduction optimization studies, especially if the cells are very sensitive or difficult-to-transduce.
Renilla Luciferase Internal Control Lentivirus – Ready-to-transduce lentiviral particles expressing minimal TATA box-driven Renilla luciferase (RLuc).
