NFκB-ASC (Pyroptosis) Reporter is designed to monitor the formation of pyroptotic ASC specks in response to inflammasome activation. This system expresses GFP-tagged ASC, enabling visualization of pyroptotic ASC specks as markers of inflammasome complex formation. ASC polymerization is triggered by danger signals, forming specks that accumulate in the extracellular space. This reporter can be stably expressed in appropriate cells, such as THP-1 cells, and is useful for studying pyroptosis, a pro-inflammatory form of programmed cell death, in vitro.
This product is supplied as high-titer lentiviral particles (third generation), which have been purified by PEG precipitation and sucrose gradient centrifugation. The lentiviral particles are VSV-G pseudotyped and are ideal for difficult-to-transfect cells, including primary or thawed cells.

Background
Pyroptosis, a form of programmed cell death, plays a critical role in the innate immune response. It is primarily triggered by inflammatory signals that activate the NLRP3 inflammasome, leading to the assembly of the ASC pyroptosome and subsequent caspase-1 activation. This process results in the formation of gasdermin D pores in the cell membrane, allowing the release of pro-inflammatory cytokines like IL-1β. Pyroptosis is implicated in various diseases, including cancer and autoimmune disorders, and targeting this pathway is a growing area of research for therapeutic development.
Vector Layout
The ASC-GFP fusion protein is expressed under the control of NFκB response elements linked to a minimal TATA promoter and an upstream enhancer. Separately, the EF1α promoter drives the expression of the puromycin selection marker, allowing for stable cell line generation. This vector design enables easy detection of NFκB pathway activation via GFP fluorescence and efficient selection with puromycin.

Figure 1. THP-1 cells (5×10^5) were transduced with 50 µl lentivirus of NF-κB-ASC-GFP-Puro and spun for 2 hours. Cells were cultured for 72 hours and selected with 1.5-2 µg/ml puromycin for 7-10 days. They were then treated with 1 µg/ml LPS for 16 hours and 2.5 µg/ml polyIC mixed with Lipofectamine for 24 hours. The extracellular ASC specks indicate pyroptotic activation. Despite being a weak inducer, intracellular polyIC also led to pyroptotic signaling.
