Non-viral ORF cDNA expression plasmids, validated for expression of the indicated gene by tag-specific antibody staining and sequencing. This is an expression vector for the human NOTCH1 N-terminal intracellular domain (NICD) which is the functional unit of NOTCH1 receptor that activates gene transcription. In full length NOTCH1, ligand binding to the extracellular portion of NOTCH1 triggers cleavage and cytosolic release of NICD, allowing NICD to enter into the nucleus and activate gene transcription. The NICD expression plasmid can be useful in establishing the correct extracellular stimulation conditions for NOTCH pathway activation in stable cell lines that have been established with the lentiviral NOTCH1 reporter product (LTV-0010). CMV drives expression of ORF cDNA fused to an N- or C-terminal epitope tag, and a downstream fluorescent reporter (GFP or RFP), separated from the cDNA by a 2A sequence. The plasmids were made with optimized vectors for high expression in mammalian cells and are supplied as transfection-ready DNA.

Applications
The NOTCH1 NICD or “cNOTCH” expression vector can be useful in helping establishing the correct extracellular stimulation conditions for NOTCH pathway activation in stable cell lines that have been transduced and selected with the NOTCH1 reporter lentivirus (LTV-0010). Please note that the transcriptional activation of NOTCH1 NICD is still dependent on its interaction with other nuclear factors such as CSL and other factors, and can also be influenced by cellular context. Therefore, the NICD overexpression plasmids are intended to provide a tool for forcing NICD expression in the cells which could help determine the extracellular conditions needed to activate NOTCH receptor and release NICD naturally. However, if other intracellular components that are required for NOTCH transcriptional activity are missing, overexpressing NICD with this product may still not provide an appropriate positive control. This will vary among different types of cells.
