These non-lentiviral plasmids were made using optimized ORF-expressing vectors listed in the vector category. The fluorescent reporter (GFP or RFP) expression is driven by CMV promoter and the reporter is either fused to ORF or separated by P2A/T2A. Supplied as transfection-ready DNA.
The addition of self-cleaving peptides between ORF and reporter allows for their separate translation using the same mRNA transcript. The ORF plasmids with P2A/T2A fluorescence reporters can be used for the quantification of silencing efficiency of shRNA or siRNA of the gene in its ORF region by co-transfection, which gives rise to a reliable, accurate result in a short amount of time. All non-lentiviral ORF plasmids were made with vectors for high expression in mammalian systems. The tags have been fused to ORF by either N-terminal (tag-ORF) or C-terminal (ORF-tag) depending on the product.
