The PD1-CD28/NFAT Immunotherapy Reporter is designed to measure T cell activity in response to modulation of the PD-1 pathway. The lentiviral construct utilizes EF1a promoter to drive constitutive expression of human PD1 and CD28, which are separated by a P2A linker, resulting in the expression of two separate proteins in the transduced cells. Downstream of CD28 is a drug selection marker or fluorescent reporter gene which is also constitutively expressed as a separate protein, allowing the user to select drug resistant or fluorescent cells for easy generation of stable cell lines. A separate expression cassette within the same construct utilizes tandem repeats of NFAT response elements coupled to a minimal TATA promoter and upstream proprietary enhancer element to drive expression of the reporter gene(s), which include a fluorescent reporter (GFP or RFP) and/or Gaussia Luciferase (GLuc), providing a convenient approach to read out T cell activity. This product is supplied as high-titer lentiviral particles, which have been purified by PEG precipitation and sucrose gradient centrifugation. The lentiviral particles are VSV-G pseudotyped and are ideal for difficult-to-transfect cells including primary or thawed cells.

Immune checkpoint inhibitor assay, T cell activation assay. The product can be used to generate stable cell lines that read out PD1/CD28 signaling through downstream activity of NFAT, for example in stable T cell lines. The fluorescent and luminescent dual reporter provides flexibility in the readout approach. GLuc is secreted, allowing the user to detect it in various biological media. Generally, the product is useful for analyzing T cell activity as a function of PD1/CD28 stimulation/inhibition. |

TF reporter products such as this one are not available in plasmid format. They are only available as high-titer lentiviral particles. Other gene products on our site, such as ORF cDNA and shRNA products are available in either format.
