The SIRPα/NF-κB Immunotherapy Reporter Lentivirus is designed to investigate the CD47-SIRPα pathway, known as the “don’t eat me” pathway, which plays a significant role in tumor immune evasion by inhibiting macrophage phagocytosis. CD47 inhibits phagocytosis by binding to signal-regulatory protein alpha (SIRPα) on phagocytes, leading to tyrosine phosphatase activation, inhibition of myosin accumulation at the phagocytic synapse, and suppression of NF-κB activation in macrophages.
CD47, a cell surface molecule in the immunoglobulin superfamily, is expressed on both normal and tumor cells and interacts with several proteins in addition to SIRPα on macrophages and dendritic cells (DCs). The SIRPα/NF-κB reporter lentivirus is an effective tool for generating stable cell lines to measure the potency of SIRPα blocking antibodies or SIRPα-Fc in counteracting tumor evasion from macrophage-mediated killing.
This product features a dual reporter system comprising secreted Gaussia luciferase (GLuc) and GFP, driven by NF-κB response elements, and is designed for high-efficiency transduction and stable expression in your cells of interest. The all-in-one high-titer purified lentivirus facilitates the generation of robust cell lines for functional assays.
This product is supplied as high-titer lentiviral particles (third generation), purified by PEG precipitation and sucrose gradient centrifugation. The lentiviral particles are VSV-G pseudotyped, ideal for transducing difficult-to-transfect cells, including primary or thawed cells.

The fluorescent and luminescent dual reporter provides flexibility in the readout approach. GLuc is secreted, allowing the user to detect it in various biological media. Generally, the product is useful for analyzing cell activity as a function of SIRPA stimulation/inhibition. |

TF reporter products such as this one are not available in plasmid format. They are only available as high-titer lentiviral particles. Other gene products on our site, such as ORF cDNA and shRNA products are available in either format.
