The STING/ISRE Immunotherapy Reporter Lentivirus is designed to evaluate the activation of the cGAS-STING pathway, a critical innate immune pathway involved in defense against viral infections and tumor growth through the induction of type I interferons (IFNs) and inflammatory cytokines. This pathway senses cytosolic double-stranded DNA (dsDNA) to activate NF-κB and IRF3, which binds to the ISRE to induce inflammatory cytokines and IFNs.
The STING/ISRE reporter is an essential tool for establishing stable, sensitive cell lines for screening cGAS-STING pathway agonists. It enables the monitoring of the effects of these agonists on the regulation of the tumor microenvironment, particularly in association with antigen-presenting cells, dendritic cells (DCs), and antitumor T cells.
This product features a dual reporter system comprising secreted Gaussia luciferase (GLuc) and GFP, driven by ISRE elements, and is designed for high-efficiency transduction and stable expression in your cells of interest. The all-in-one high-titer purified lentivirus facilitates the generation of robust cell lines for functional assays.
This product is supplied as high-titer lentiviral particles (third generation), purified by PEG precipitation and sucrose gradient centrifugation. The lentiviral particles are VSV-G pseudotyped, ideal for transducing difficult-to-transfect cells, including primary or thawed cells.

STING/ISRE Assay. The product can be used to generate stable cell lines that read out STING signaling through downstream activity of ISRE. The fluorescent and luminescent dual reporter provides flexibility in the readout approach. GLuc is secreted, allowing the user to detect it in various biological media. Generally, the product is useful for analyzing cell activity as a function of STING stimulation/inhibition. |
*Transduction units (TU), based on the number of drug resistant or fluorescent HEK293FT cells after transduction with the virus.
