  • Versatile readout of gene expression – Our ORF cDNA constructs are New-Version Vectors that have been optimized for production of high-titer lentiviral particles and easy establishment of stable cell lines. All of our ORF cDNA constructs feature an epitope tag (e.g. Flag, V5, Myc, HA, etc.) fused to the ORF C-terminus, allowing easy purification or staining of fixed cells/lysates. In live cells, drug selection markers allow stable cell line generation in ~10 days; or, fluorescent reporter can allow isolation by FACS.  Track cells in vivo or in vitro using fluorescence or luciferase. Self-cleaving peptides are included to allow independent protein expression of these components from a single mRNA transcript.
  • High-titer & ultra purifiedLipExoGen  lentiviral particles are ultra-purified and concentrated to high-titer by PEG precipitation and sucrose gradient centrifugation. For immunotherapy/functional assay reporter lentiviruses, if the receptor cDNA ORF exceeds 1.2 kb in the all-in-one dual-reporter construct (e.g. GFP-GLuc), transduction efficiency will be largely reduced, due to it exceeds our packaging capacity. In such cases, we provide the cDNA ORF lentivirus and the reporter lentivirus as a set instead. You can use this set for co-transfection, ensuring high-efficiency transduction without additional time delays. The price for this set is $1200.
If you need a cDNA expression plasmid that isn’t listed in the table, you may request any cDNA sequence. For cDNA size under 2.5 kb, pricing will be similar to the listed options. Larger cDNAs may incur additional costs. You can also specify your preferred vector, and the price will remain comparable for plasmids with cDNAs under 2.5 kb. Self-cleaving peptide shorthand: T2A and P2A sequences are denoted using slashes (“/”) in ORF cDNA product names. Example: Gene-V5/Reporter/SelectionMarker represents Gene-V5–T2A–Reporter–P2A–SelectionMarker. We use the “-” symbol to indicate fusions. Example: “Gene-Tag” means the tag is fused to the C-terminal of the ORF cDNA, while “Tag-” means the tag is fused to the N-terminal. You can view the diagram of the construct layout here.
